簡(jiǎn)介:南京農(nóng)業(yè)大學(xué)博士學(xué)位論文RXLR效應(yīng)分子協(xié)同互作控制大豆疫霉對(duì)寄生侵染過(guò)程姓名王群青申請(qǐng)學(xué)位級(jí)別博士專業(yè)植物病理學(xué)指導(dǎo)教師王源超201007博士學(xué)位論文RXI,R效應(yīng)分子協(xié)同互作控制大豆疫霉對(duì)寄主的侵染過(guò)程術(shù)干擾兩類表達(dá)量較高的效應(yīng)分子中的關(guān)鍵基因肭刀8和彳砌J『力的轉(zhuǎn)錄水平,破壞其轉(zhuǎn)錄表達(dá)模式,則導(dǎo)致菌株致病力明顯下降。說(shuō)明這種程式化的轉(zhuǎn)錄模式一旦發(fā)生紊亂,就會(huì)影響菌株的毒性,同時(shí)也證明兩類高表達(dá)的效應(yīng)分子在侵染過(guò)程中都具有重要作用。對(duì)不同表達(dá)模式的效應(yīng)分子的功能做進(jìn)一步研究分析發(fā)現(xiàn),E類的效應(yīng)分子能夠特異J生抑制P秈觸發(fā)的植物免疫反應(yīng),而IE類效應(yīng)分子則特別對(duì)效應(yīng)分子觸發(fā)的PCD有活性。兩類效應(yīng)分子分別針對(duì)植物免疫系統(tǒng)的不同層面,推測(cè)可能合作抑制植物防衛(wèi)反應(yīng)。本研究在煙草上成功模擬了兩類效應(yīng)分子的合作模式卵菌的啪被植物的膜上受體識(shí)別激發(fā)PTI,誘導(dǎo)E類效應(yīng)分子如AVLL238在侵染前期高量上調(diào)表達(dá),引起植物包括非寄主煙草和寄主大豆的PCD,而在侵染過(guò)程中一直表達(dá)量較高的AVHL72則能夠抑制AVH238引起的PCD,而兩者的同時(shí)存在還能夠抑制斟F1以及MAPK信號(hào)通路在煙草上引發(fā)的過(guò)敏性細(xì)胞死亡,證明兩類效應(yīng)分子能夠依靠功能互補(bǔ)來(lái)廣泛抑制植物的防衛(wèi)反應(yīng)本研究還發(fā)現(xiàn)砒乃8在不同菌株中的等位基因存在著高度的序列多態(tài)性其中來(lái)自菌株P(guān)7076的彳訛2始等位基因產(chǎn)物在煙草中的表達(dá)產(chǎn)物不能引起HR功能實(shí)驗(yàn)則證明AVH238P7076能夠抑制在煙草葉片上瞬時(shí)表達(dá)卵茵N執(zhí)口INFL以及下游的MAPK信號(hào)通路所引起的胍反應(yīng)。證明這種序列的多態(tài)性能夠幫助RXLR效應(yīng)分子逃避植物對(duì)其的識(shí)別作用,但不會(huì)影響其本身的毒性功能。對(duì)所有已知的無(wú)毒基因和兩類在侵染中高表達(dá)的效應(yīng)分子序列進(jìn)行分析比較也發(fā)現(xiàn),這種序列多態(tài)性在RXLR家族成員中普遍存在,說(shuō)明其作為卵茵直接與植物互作的蛋白,面臨著巨大的選擇壓力,所以進(jìn)化速度非???。而對(duì)觸地38的進(jìn)一步研究發(fā)現(xiàn),其蛋白序列笫51及第76位氨基酸與其觸發(fā)植物免疫反應(yīng)顯著相關(guān),但這兩個(gè)氨基酸并不位于已知的功能域上。說(shuō)明這種序列的多態(tài)性能夠幫助RXLR效應(yīng)分子逃避植物對(duì)其的識(shí)別作用,但不會(huì)影響其本身的毒性功能。關(guān)鍵詞大豆疫霉;無(wú)毒基因;I乩R效應(yīng)分子;毒性功能;表述模式;多態(tài)性Ⅱ

簡(jiǎn)介:我國(guó)是一個(gè)丘陵和山區(qū)占很大面積的農(nóng)業(yè)大國(guó),類似微耕機(jī)這樣的小型機(jī)械已經(jīng)得到了廣泛的應(yīng)用。隨著農(nóng)業(yè)農(nóng)村改革的不斷深入、種植結(jié)構(gòu)的不斷調(diào)整,大田作物面積不斷減少,經(jīng)濟(jì)作物面積逐步增加,品種種植也越來(lái)越趨向于規(guī)?;?,如何降低耕作勞動(dòng)強(qiáng)度、實(shí)現(xiàn)耕作無(wú)人化已逐漸成為人們關(guān)注的熱點(diǎn)。結(jié)合日益成熟的無(wú)線遙控技術(shù),研制出低成本、高性能的微耕機(jī)遠(yuǎn)程控制系統(tǒng),是本課題的主要研究目的。論文從微耕機(jī)遠(yuǎn)程控制系統(tǒng)技術(shù)方案的理論依據(jù)出發(fā),詳細(xì)介紹了無(wú)線遙控技術(shù)的相關(guān)知識(shí)和系統(tǒng)需求分析及方案論證,規(guī)劃了微耕機(jī)遠(yuǎn)程控制系統(tǒng)的總體設(shè)計(jì)思路。最后確定采用低功耗無(wú)線技術(shù)作為無(wú)線傳輸方式和以高性能ARM微控制器LPC2132為主控芯片的方案,并給出了系統(tǒng)各個(gè)組成單元的功能、軟硬件結(jié)構(gòu)、通信協(xié)議等,使用模塊化的架構(gòu)簡(jiǎn)化了系統(tǒng)的設(shè)計(jì),為后面的設(shè)計(jì)與試驗(yàn)奠定了基礎(chǔ)。論文設(shè)計(jì)了系統(tǒng)的硬件電路,硬件實(shí)現(xiàn)選用ARM系列微處理器LPC2132作為核心控制芯片,采用無(wú)線傳輸模塊NRF905實(shí)現(xiàn)數(shù)據(jù)的發(fā)送和接收,使用液晶顯示模塊LCD12864來(lái)實(shí)時(shí)顯示系統(tǒng)的運(yùn)行狀態(tài)。論文以ADS12軟件為開發(fā)環(huán)境,采用C語(yǔ)言進(jìn)行編寫,以模塊化的方式,給出了控制系統(tǒng)的主程序、菜單程序、通信程序、顯示程序和中斷程序流程框圖。此外,本文還對(duì)系統(tǒng)進(jìn)行了詳細(xì)的軟、硬件抗干擾設(shè)計(jì),全面提升了系統(tǒng)的穩(wěn)定性。實(shí)驗(yàn)測(cè)試驗(yàn)證表明,本系統(tǒng)運(yùn)行可靠,工作正常、控制自如、顯示及時(shí),滿足系統(tǒng)性能的需求,有利于降低人力耕作的勞動(dòng)強(qiáng)度,實(shí)現(xiàn)了微耕機(jī)耕作無(wú)人化的目標(biāo)。
